Why Bispecific Targeting Matters
Single-antigen CD19 CAR-T fails in 30β50% of patients due to antigen escape β cancer cells lose CD19 and become invisible. Targeting CD19 and CD22 simultaneously makes escape much harder.
CD19 Antigen Escape
CD19-negative relapse occurs in 30β50% of patients after CD19 CAR-T. The cancer mutates to stop expressing CD19 β making the therapy blind to it.
CD22 as a Complementary Target
CD22 is expressed on most B-cell cancers independently of CD19. Targeting both requires the cancer to lose two antigens simultaneously to escape β a much rarer event.
