The Core Idea Behind MCTL

Instead of targeting one cancer marker, MCTL trains T cells against a personalised combination of tumour-associated antigen peptides โ€” reducing the risk of antigen-escape resistance common in single-target approaches.

โ€œA curated target library means the therapy can be personalised to each patient's tumour antigen profile rather than applying a fixed single target to all patients.โ€
  • Multi-Antigen Targeting

T cells are trained against several tumour-associated antigens โ€” such as MAGE family, survivin, WT1, NY-ESO-1, and others โ€” drawn from a curated reference library matched to the patient's tumour profile.

  • Dendritic Cell Bridge

Autologous dendritic cells are pulsed with the selected peptide combination. These cells act as antigen-presenting vehicles that activate and expand tumour-specific T cells in the laboratory.

How Bohai Biotech MCTL Is Manufactured

The manufacturing process is personalised to each patient's antigen profile.

  1. 1

    Tumour Antigen Profiling

Pathology and biomarker review to identify which tumour-associated antigens from the reference library are expressed in the patient's cancer.

  2. 2

    Leukapheresis

Peripheral blood drawn; T cells and monocytes (which become dendritic cells) are separated.

  3. 3

Dendritic Cell Preparation & Peptide Pulsing

Monocytes are matured into dendritic cells and loaded with the personalised combination of tumour-associated antigen peptides.

  4. 4

    T-Cell Expansion

Dendritic cells are co-cultured with the patient's T cells, activating and expanding tumour-specific cytotoxic T lymphocytes to therapeutic numbers.

  5. 5

    Reinfusion

Expanded MCTL cells are quality-tested and reinfused, often in combination with toripalimab.

The Published Clinical Evidence

The key published evidence for this platform is a Phase Ib trial in advanced NSCLC combining MCTL with toripalimab.

  • Phase Ib Trial: MCTL + Toripalimab in Advanced NSCLC

Published in Cancer Immunology, Immunotherapy (2026). 21 patients with advanced NSCLC received personalised peptide-pulsed dendritic cell-based MCTL combined with toripalimab as second-line therapy. The study reported manageable safety and encouraging early activity. It was single-arm and small โ€” results require confirmation in larger studies.

  • What the Evidence Does and Does Not Show

Early feasibility, acceptable safety, and encouraging disease control signals. It does not confirm superiority over standard care in a randomised trial. Patients should understand this distinction before making treatment decisions.
